Standard droplet microfluidics has enabled remarkable single-cell work — but the closed nature of conventional droplets has always imposed a ceiling on what workflows can achieve. Cells and reagents go in together, and nothing changes after that. Every complex multi-step protocol is a workaround for this constraint.
